Sanger sequencing, also known as dideoxy sequencing or chain termination sequencing, is a widely used method for DNA sequencing. Developed by Frederick Sanger and his colleagues in 1977, it has played a pivotal role in advancing molecular biology and genomics. Sanger sequencing is still routinely used for various applications, including DNA sequencing for plasmids, PCR products, and verification of cloned genes. However, for large-scale genomic projects, NGS technologies are more commonly employed due to their higher throughput and cost-effectiveness.

Sample Requirements:

To ensure proper quality of template DNA/ PCR product, the client is responsible to check samples by running gel electrophoresis prior to shipping. A single clear DNA band must be detected when loading the sample on 0.8-1.0% agarose gel (for genomic DNA) and 2% for PCR product using EtBr/SYBR Safe. Smear, multiple-bands and primer dimers for PCR product may critically affect the quality of Sanger sequencing results. Genomic DNA templates should be provided at the concentrations of 10-50ng (20.0µl) along with the primers having concentration of 10uM (10µl). Moreover, PCR product bands should be clearly visible on Gel image.
NOTE: The UV absorbance or spectrophotometry provides the quantity of DNA but doesn’t ensure the quality of the genomic DNA or PCR product.

Company takes the responsibility of providing good quality sequencing results based on passing the quality standards of PCR product or genomic DNA provided by the customers. Based on our Company experience, the sample quality mentioned above provides good quality sequencing results. However, samples can be run on sequencer upon customer’s request after taking the consent. No re-sequencing of samples shall be entertained if the error arises from customer’s side.

Company provides DNA sequencing results electronically in raw data files, pdf and ab1 files to the customers. The sequencing results can be archived in the Laboratory data base up to 6 months. During this period, customer may request sequencing results. After 6 months, DNA sequencing results will be automatically deleted without further notification. For Standard Sanger Sequencing, TAT is 7-10 days from the time the client’s samples have been received by the Company. Any additional services such as DNA extraction, PCR amplification, may delay TAT up to a week.

Samples will be discarded after 15 days upon arrival at the Company sequencing facility. If the client wishes to obtain the remaining volume of the samples shipped back to his/her location after completion of each batch, the request should be made no later than 15 days after the completion of respective batch. Shipment cost shall be beard by the Customer.
